2,4-D和KT对大花金挖耳愈伤组织诱导的影响 被引量:2

摘要 以大花金挖耳无菌苗的根为外植体,研究了不同质量浓度的2,4-D和KT对愈伤组织诱导的影响。结果表明:低浓度的2,4-D(0.25~0.5mg/L)单独使用促进愈伤组织形成,随着2,4-D质量浓度的增加,愈伤组织的形成逐渐变弱;在培养基、2,4-D和KT诱导愈伤组织的正交试验中,发现2,4-D对愈伤组织诱导率的影响最大,基本培养基次之,KT影响最小,对愈伤组织诱导的较优组合为B2+2,4-D0.1mg/L+KT0.4mg/L;在优化诱导愈伤组织的激素浓度配比试验中发现B5+2,4-D0.1mg/L+KT0.2~0.4mg/L对大花金挖耳愈伤组织的诱导率为100%,且产生的愈伤组织多为淡黄色、质地疏松、生长快、褐化时间长,适合于大花金挖耳愈伤组织的诱导。 This experiment inspected the effect of 2,4-D and KT on callus induction of Carpesium macrocephalum Franch. et Sav by taking its root as the explants from its seeds in this paper. The results showed that the low concentration 2,4-D could promote callus formation ,the frequency of callus was decreased with increased 2,4-D concentration in medium. It also showed that such factors as basic media,types and concentration of hormones had a great effect on the induction rate , growth and browning time of the callus. The proper callus induction medium was selected out by orthogonal design, more over, the kind and suitable level of hormone were ascertained. The results showed that 2,4- D was the critical factor to callus induction,then basic medias, the optimum medium for callus induc tion 6f Carpesium macrocephalum Franch. et Sav was B5 +2,4-D 0.1 mg/L+KT 0.4 mg/L. Moreover the best effects were obtained by using concentrations of B5+2,4-D 0.1 mg/L +KT0.2-0.4 mg/L in B5 solid mediums at the light of 2 000-3 000 lx ,at temperature with 25±2℃ and pH 5.8. The induction rates of the callus were 100% to its root, the callus was primrose yellow and loose,the brown ing time was late.
