杜仲腰痛丸对髓核非压迫性突出大鼠背根节中降钙素基因相关肽含量的影响 被引量:12

Effects of Duzhongyaotong pill on calcitonin gene related peptide in non-compressive nucleus pulposus protrusion in rat dorsal root ganglion

摘要 目的观察杜仲腰痛丸对髓核非压迫性突出大鼠背根节(DRG)中降钙素基因相关肽(CGRP)含量的影响。方法取健康、雄性Wistar大鼠50只,随机分为假手术组(A组)、模型组(B组)、阳性对照组(C组,伸筋丹组)、杜仲腰痛丸高剂量组(D组)、杜仲腰痛丸低剂量组(E组),每组10只。将大鼠自身的尾椎髓核取出移植于左侧L5和L6神经根背侧,造成大鼠非压迫性髓核突出模型,通过免疫组化的方法测定大鼠2周时DRG中CGRP的变化。结果B、C、D和E组DRG中CGRP阳性神经元的表达明显增加,B、C、D和E组CGRP阳性神经元的免疫反应强度与A组比较均增高(P<0.05),其中B组与A组比较显著增高(P<0.01),但C、D、E组与B组比较为低(P<0.05),D、E组与C组比较为低(P<0.05)。结论杜仲腰痛丸可通过下调或者抑制非机械压迫性髓核对神经根损伤后DRG内的痛觉神经递质CGRP的释放而发挥镇痛作用。 Objective To observe the effects of duzhongyaotong pill on calcitonin gene related peptide(CGRP) in non-compressive nucleus pulposus protrusion in rat dorsal root ganglion(DRG). Methods 50 normal Wistar male rats were randomly divided into five groups: sham operated group(A), injury model group(B), shenjindan capsule treated group(C), high and low dose duzhongyaotong pill treated groups(D and E) (n = 10 for each group). The model of non- compressive nucleus pulposus protrusion was established by transplanting autogenous nucleus pulposus in the back of left L5-L6 nerve roots. CGRP in DRG was determined by immunohistochemistry at two weeks. Results The number of CGRP positive neurons in DRG of B, C, D and E groups increased significantly, moreover, the intensity of immunoreaction of CGRP positive neurons was stronger in B, C, D and E groups than in A group(P 〈 0.05), and it was significantly stronger in B group than in Agroup(P 〈 0.01), but it was lower in C, D and E groups than in B group(P 〈 0.05), and lower in D and E groups than in C group(P 〈 0.05). Conclusion The analgesic mechanism of duzhonhyaotong pill may be by down-regulating the expression of algesia neurotransmitter CGRP in neurons of DRG in non-compressive nucleus pulposus protrusion nerve root injury.
