长期高脂饲养对大鼠胰岛细胞功能和凋亡的影响及玉液汤化裁的干预作用 被引量:3

摘要 目的观察长期高脂饲养对大鼠胰岛细胞凋亡的影响及玉液汤化裁对脂毒性的防治作用.方法 SD 8周龄大鼠24只,随机分为正常饲料组(NC,n=6)、高脂饲料组(HF,n=6)、高脂饲料加二甲双胍干预组(MT,n=6)及高脂饲料加玉液汤干预组(HM,n=6),不同饲料喂养6个月,行口服葡萄糖耐量试验及胰岛素释放实验,测血游离脂肪酸(FFAs).实验结束时,处死大鼠,计量内脏脂肪/体重比值,胰腺切片用HE染色、免疫组化和TUNEL方法观察各组胰岛形态学变化.结果实验结束时, 0~60 min血糖曲线下面积(AUCG)HF组均极显著大于NC组、MT组和HM组(P<0.01);0~120 min HF组均极显著大于NC组、MT组和HM组(P<0.01).HF组空腹血游离脂肪酸显著高于NC组(P<0.01),HM组和MT组FFAs较HF组有明显降低趋势.胰岛细胞凋亡百分率HF组显著高于NC组(P《0.01),HM组和MT组均显著低于HF组(P<0.01).结论长期高脂饲养正常SD大鼠可引起胰岛细胞凋亡,玉液汤化裁在一定程度上可对抗脂毒性. Objective To investigate the effect of long-term high-fat feeding on apoptosis rate of pancreatic island( cells in rats and intervention effects of Yuyetanghuacai(YYTH, a compound herbal recipe). Methods Twenty four healthy male Sprague-Dawley (SD) rats, 8 weeks of age,were randomly divided into 4 groups: normal control group (NC, n =6), high fat diet group (HF, n = 6) , herbal medicine intervention group (HM, n = 6) , metformin intervention group ( MT, n =6). The animals were subjected to an oral glucose tolerance test at the age of 32 weeks, pancreatic tissues slices were stained by and terminal-dcoxynueleotidyl transferase mediated nick end labeling (TUNEL) methods ReaCts At the end of the experiment, plasma fasting free fatty acids (FFFAs) concentration in HF group was significantly higher than that in NC group( P 〈 0.01 ). The percentage of islet cells apoptosis in HF group was significantly higher than that in NC group( P 〈 0.001 ). The percentage of islet cells apoptosis in HM group and MT group were significantly lower than that of HF group( P 〈 0.01, P 〈 0.001 respec.tively). Conclusion A long-term high-fat feeding results in increase of blood FFAs and the increase of apoptosis rate of islet cells in SD rats. YYTH can prevent lipotoxieity at some extents.
