单侧输尿管结扎致大鼠肾间质纤维化模型中CTGF、α-SMA的动态观察 被引量:2

摘要 目的:对肾脏结缔组织生长因子(CTGF)、α平滑肌肌动蛋白(α-SMA)在单侧输尿管结扎致大鼠肾间质纤维化模型中的表达进行动态观察,以探讨CTGF在肾间质纤维化病程中的作用及意义。方法:采用单侧输尿管结扎术(UUO)致大鼠肾间质纤维化模型,分别于术后第1、3、5、7、9、12天处死大鼠,HE、Mmson染色观察病理改变并进行小管间质损伤指数评分,免疫组织化学方法观察各组大鼠CTGF、α-SMA的表达。结果:第1~9天大鼠肾间质纤维化病理改变逐渐明显,CTGF、α-SMA表达逐渐增多(各组间比较,P〈0.05);CTGF与肾小管间质损伤指数、甜SMA为正相关关系。结论:CTGF在肾小管间质纤维化的病程进展中起重要的作用,并可作为评价肾纤维化病变程度的良好指标及防治的新的靶目标。 Objective: To continuously observe the expression of CTGF and α-SMA in renal tubulointerstitial fibrosis of rats, and to investigate the significance of fibrogenic effects of CTGF. Methods: Unilateral ureteral obstruction (UUO) was adopted to produce rat models of renal tubulointerstitial fibrosis, and the rats were executed on the 1^st, 3^rd, 5^th, 7^th, 9^th and 12^th days post-operation respectively. The expression of CTGF,α-SMA in the obstructed kidney were measured by immunohistochemistry. At the same time, the degree of tubulointerstitial damage was scored. Results: The levels of CTGF, α-SMA increased gradually from the first day to the 9th day (P 〈 0.01 ) and paralleled the degree of tubulointerstitial damage. CTGF had positive correlation with the indexes of tubulointerstitial damage and α-SMA. Conclusions: CTGF may play an important role in the development of tubulointerstitial fibrosis and become a new cure target for the prevention of the disease.
