自体骨髓细胞移植对老年猕猴肝脏功能与体液免疫及血细胞比例的干预

摘要 目的:分析自体骨髓细胞肝门静脉植入后对老年灵长类动物肝脏功能、体液免疫及血液细胞亚群数量变化的影响。方法:实验于2004-04/12在昆明总医院医学实验科完成。选取18~20岁老年恒河猴8只,分离骨髓有核细胞,将3×109个自体骨髓单个核细胞经肝门静脉植入体内。于骨髓细胞移植前和移植后1,2,3,4,5,6个月分别采集外周血5mL,吸取上清液采用自动生化分析仪观察外周血肝功相关蛋白含量、胆红素含量及肝酶活性的变化;以上各时间点另采集外周血2mL,应用SF-300全自动血球分析仪测定外周血白细胞总数、淋巴细胞、粒细胞、单核细胞、血小板及红细胞的数量和比例。结果:实验选用8只恒河猴,全部进入结果分析。①骨髓细胞移植后外周血肝功相关蛋白含量及肝酶活性的变化:与骨髓细胞移植前比较,血清总蛋白、清蛋白及球蛋白含量在移植后6个月内均明显升高(P<0.05);谷丙转氨酶、血清谷草转氨酶活性在移植后1,2,3个月内明显升高(P<0.05),随后逐渐下降,至6个月时接近移植前水平(P>0.05)。②骨髓细胞移植后血清胆红素浓度的变化:与骨髓细胞移植前比较,至移植后6个月血清总胆红素浓度明显降低(P<0.05),血清间接胆红素浓度与移植前基本相似(P>0.05);血清直接胆红素浓度在细胞移植后6个月内一直低于移植前水平(P<0.05)。③骨髓细胞移植后血清免疫球蛋白浓度及外周血细胞亚群比例的变化:血清IgG,IgA,IgM,C3,C4浓度、白细胞总数、血小板数量和淋巴细胞、单核细胞、粒细胞比例在移植后6个月内均显著高于细胞移植前(P<0.05);红细胞数量未见明显变化。结论:老年猕猴自体骨髓细胞肝门静脉移植能够改善肝脏的蛋白合成和代谢功能,具有增强细胞与体液免疫力的作用。 Abstract AIM: To analyze the effect of transplantation of autologous bone marrow through hepatic vein on liver function, humoral immunity and the number of hematological cell-subgroups in primate. METHODS:This experiment was carried out at the Department of Laboratory Medicine, Kunming General Hospital from April to December' 2004. Eight rhesuses , aged 18 to 20 years, were used in this experiment. Karyotes were isolated from the bone marrow, 3 ×10^9 autologous mononuclear bone marrow cells were transplanted into the body through hepatic vein. A total of 5 mL peripheral blood was collected before transplantation of bone marrow cells and at 1,2,3,4,5,6 months after transplantation. Supernatant liquid was extracted to observe the levd change of liver function related protein, bilirubin and enzyme activity of liver;, Another 2 mL peripheral blood was collected at each time point, then SF-300 automatic aloed biochemical analyzer was applied to measure the number of leukocyte, lymphocyte, granulocyte, monocyte, platelet and erythrocyte in peripheral blood as well as their ratios. RESULTS: Eight rhesuses were used in this experiment, and all of them entered the stage of result analysis. (1)Change of liver function related protein content and enzyme activity of the liver:. As compared with that before transplantation, the level of total serum protein, albumen and globulin was obviously increased within 6 months after transplantation (P 〈 0.05); The activities of glutamate-pyruvate transaminase and serum glutamic-oxaloacetic transaminase were significantly increased within 1, 2 and 3 months after transplantation(P 〈 0.05), then decreased gradually , and close to the level at the 6^th month after transplantation (P 〉 0.05). (2) The level of serum total bilirubin was significantly decreased at the 6^th month after transplantation as compared with that before transplantation (P 〈 0.05). Indirect level of serum bilirubin was basically similar to that before transplantation (P 〉 0.05);Direct level of serum bilirubin was lower within 6 months after transplantation than that before transplantation (P 〈 0.05).(3) The level of serum IgG,IgA,IgM,C3,C4 in serum and the ratio of total white blood cells, platelet , lymphocyte, monocytes and granulocytes were all significantly higher within 6 months after transplantation than before transplantation (P 〈 0.05); There was no obvious change of the number of akaryocyte. CONCLUSION: Autognlous bone marrow cells transplantation through hepatic vein of old rhesus can improve the protein synthesis and metabolism function of liver and strengthen the cellular and humoral immunity.
