ADPRT抑制剂3—氨基苯甲酰胺对博来霉素的毒性及抗瘤作用的影响

摘要 本实验发现ADPRT抑制剂3 AB2.5mmol/kg与BLM2.5mg/kg联合用药,便BLM对S180的抑瘤率从42.5%、46.1%提高到66.2%、75.9%,而小鼠体霞无明显减轻。BLM 5mg/kg单独用药,抑瘤率为64.8%、75%、抗瘤作用与上述联合用药组相当-但其体重减轻更为明显。3AB在有协同抗瘤作用的剂量下,对BLM的急性毒性无增毒作用。在亚急性毒性方面,通过观察小鼠体重变化、外周血白细胞计数、肺形态学表现及肺湿重、羟脯氨酸含量,发现3 AB对BLM的毒性无增强作用。实验为更有效地将无明显毒性的ADPRT抑制剂与BLM联合用药治疗肿瘤提供了实验依据。 In this experiment it was shown that the combined use of 3AB 2.5mm-ol/kg and BleomyCin (BLM) 2.5mg/kg can increase the inhibition rate of BLM atone against S-180 from 42.5 and 46.1% to 66.2 and 75-9% ; while the reduction of body weight of mice was not increased significantly. BLM 5mg/kg used alone has induced inhibition rates of 64.8 and 75% against S-180, similar to those observed in the combination groups, but the reduction of body weight was more significantly.The acute toxicity of BLM was not enhanced by the combination of 3AB B 2.5mmol/kg,under this dosage the antitumor effect of BILM was enhanced. In the aspect of subacute toxicity, through the observations on the changes of body weight, peripheral blood white cell count, histological and ultrastructural changes of the lung, wet weight of the lung and the hydroxyproline content of the lung, it was shown that there was no significant difference between the BLM alone group and BLM + 3AB group.This paper has Provided experimental basis for the effective combination of BLM with low toxic drug (ADPRT inhibitor. 3AB) in cancer chemotherapy.
