摘要
目的探讨表皮生长因子(EGF)对人舌鳞癌细胞株基质金属蛋白酶(MMP)-7表达的影响及其机制。方法对体外培养的人舌鳞癌细胞株用重组人表皮生长因子(rhEGF)作为刺激因子,采用免疫组化SP法、半定量RT-PCR和Westernblot方法测定癌细胞中MMP-7mRNA和蛋白含量变化。结果与未处理组相比,用不同浓度(0.1、1.0、10.0ng/ml)EGF处理癌细胞后MMP-7mRNA和蛋白表达随EGF浓度的增加而增加,酪氨酸激酶抑制剂Genistein(10μg/ml)能够完全抑制EGF对癌细胞MMP-7表达的诱导作用。结论EGF通过酪氨酸激酶通路诱导人舌鳞癌细胞株MMP-7的表达。
Objective To investigate the effect of matrix metalloproteinase-7(MMP-7) and its mechanism in tongue squamous cell carcinoma cell line induced by EGF. Methods The effects of rhEGF on the production of MMP-7 in tongue squamous cell carcinoma cell were stud- ied by immunohistochemistry, semiquantitative RT-PCR and Western blot assay. Results The exposure of tongue squamous cell carcinoma cells to EGF at the indicated concentrations(0.1 ng/ml, 1.0 ng/ml, 10.0 ng/ml)raised secretion of MMP-7 in a concentration-dependent manner compared with the untreated controls. We have also found the tyrosine kinase inhibitor genistein ( 10 μg/ml) totally inhibited the increased production of MMP-7 resulting from stimulation with EGF( 10 ng/ml). Condusion EGF may be involved in the production of MMP-7 via tyrosine kinase signal pathway.
