骨髓干细胞移植对实验性大鼠急性心肌梗死的影响 被引量:1

Effect of Transplanting Bone Marrow Mesenchymal Stem Cells on Pathologic Change of Rats with Acute Myocardial Infarction

摘要 [目的]通过同种异体大鼠骨髓干细胞(BMSCs)移植,观察其对实验性大鼠急性心肌梗死面积的影响。[方法]清洁级Wistar雄性大鼠无菌分离骨髓干细胞;将36只同种雄性大鼠随机分为模型组、对照组和细胞移植组。结扎大鼠左冠状动脉前降支建立急性心肌梗死模型。细胞移植组在心肌梗死周边分四点注射BMSCs细胞悬液,对照组心肌梗死区周边注射等容积BM SCs完全培养液。通过心肌组织硝基四氮唑蓝(N-BT)染色,血中血清肌酸激酶(CK)、乳酸脱氢酶(LDH)、超氧化物歧化酶(SO D)活性、丙二醛(MDA)水平检测,观察BM SCs移植对上述各指标的影响。[结果]细胞移植组能减少心肌梗死面积,降低血中CK、LDH、M DA水平,提高血中SOD水平。[结论]BM SCs移植对结扎大鼠左冠状动脉前降支所致的急性心肌梗死具有保护作用。 [Objective] To explore the effect of homogenous and heterologous transplantation of hone marrow mesenchymal stem cells(BMSCs) on rats with acute myocardial infarction. [Methods] BMSCs were separated with asepsis from female Wistar rats of clean grade. The myocardial infarction was induced in 50 homogenous male rats by ligating the left anterior descending coronary artery. 36 rats with myocardial infarction were randomly divided into model group, control group, TCM-treating group and cellular transplanting group. Isovolumic NS was injected into the parainfarcted area in control group and the BMSCs cell suspension injected at 4 points of parainfarcted area in transplantation group. The heart was isolated four weeks later for pathological examination and determination of the area of myocardial infarction. [Results] The myocardial infarction area in cellular transplant group was less than that of control group(P〈0.01). [Conclusion] The transplantation of BMSCs may decrease the infarcted size in rats with acute myocardial infarction.
