摘要
目的探讨苦参碱对大鼠原位肝移植供肝冷保存再灌注损伤中枯否细胞的影响及其作用机制。方法本实验应用延长保存的大鼠原位肝移植模型,将160只大鼠随机分为对照组、苦参碱小剂量治疗组、大剂量治疗组和假手术组,分别检验移植术后血中内毒素和肝组织中肿瘤坏死因子-α(TNF-α)的含量,并观察移植肝脏病理形态学的改变。结果与对照组比较,治疗组术后各时点肝组织内的TNF-α的含量均明显降低,内毒素血症明显减轻,枯否细胞增生活跃程度明显减轻,同时肝细胞和肝内皮细胞损伤的病理表现也明显改善。结论苦参碱可以通过减轻再灌注后内毒素治疗,有效抑制枯否细胞激活及释放TNF-α等炎症性损伤介质,保护肝细胞及肝窦内皮细胞,从而减轻供肝冷保存再灌注损伤。
Objective To study the effect of matrine on Kupffer cell activation in cold preservationreperfusion injury in rat orthotopic liver transplantation and clearify its possible mechanism. Methods One hundred and sixty Wistar rats were randomly divided into 4 groups: untreated group ( control group), lowdose treated group, high- dose treated group and sham operation group. The content of endotoxin in blood, the content of TNF - α and the pathology morphologic change in liver were detected after the long - time delayed cold preservation in orthotopic liver transplantation model of rats. Results Compared to control group, the content of endotoxin in blood, the content of TNF - α, the activation and proliferation of Kupffer cell in liver signif- icently decreased. And the pathological change of hepatic tissue and the harm of SEC were reduced greatly. Conclusion Matrine can prevent SEC and hepatic cells from cold ischemia reperfusion injury by inhibited the activation of Kupffer cell and decreased the release of TNF - α, endotoxin in blood and hepatic tissue.
