罗格列酮在Beagle犬体内血药浓度测定方法研究

摘要 目的建立高效液相色谱法测定Beagle犬体内罗格列酮血药浓度的方法。方法以十八烷基键合硅胶柱为固定相.以0.01mol/L的磷酸二氢钾水溶液-乙腈(50:50),以三乙胺调节pH6.0为流动相,检测波长247nm。4-硝基-2-苯氧基甲烷磺酰苯胺为内标,样品用乙醚提取,按内标法定量。结果标准曲线50—5000ng/ml范围内有良好线性关系(r=0.9996),最低检测浓度为30ng/ml,平均方法回收率为97.34%(RSD为2.15%)。结论本文建立的方法能够准确测定罗格列酮的血药浓度,线性关系好,回收率高,稳定耐用。 Objective To establish a assay method for the determination of rosiglitazone in serum of Beagle dogs. Method Analysis was conducted with the column of C8 (250 × 4.6 mm, 5μm) and mobile phase of 0.01 mol/L potassium dihydrophosphate-acetonitrile (50: 50) (pH value was adjusted to 6.0 with triethylamine), and rosiglitazone was determined at the wavelength of 247 nm. , and 4-nitro-2-phenoxymethane sulfonanilide as interal standard. Results In the eoneentration range of 50~5000 ng·ml^-1 , the distribution of rosiglitazone is linear (r = 0.9996) , with an average reeovery of 97.34% (RSD was 2.15% ). Conelusion The assay method is convenient, aeeurate, and reliable with good preeision, and ean be used in the determination of rosiglitazone in serum of beagle dog.
