摘要
目的:观察T细胞对大鼠胰岛瘤细胞系RINm5f凋亡及凋亡相关基因表达的影响。方法:应用PI法检测T细胞与RINm5f共培养后RINm5f细胞的凋亡率;应用半定量RT-PCR技术检测培养后bcl-2、bax、p53和fasmRNA的表达。结果:PHA与T细胞均可致胰岛细胞的凋亡,且P、T具有协同效应,P或T或两者联合应用均呈时间依赖性地促进RINm5f细胞的凋亡;胰岛细胞凋亡过程中,P、T可协同致诱导凋亡基因bax、fasmRNA表达水平明显升高,而抗凋亡基因bcl-2及p53mRNA表达水平有下降趋势,但统计学无显著性差异。结论:T细胞可通过诱导胰岛细胞凋亡导致或加重胰岛移植排斥反应,其机制可能与凋亡基因与抗凋亡基因的表达比率变化有关,其中fas、bax诱导凋亡基因可能起主要作用。
Objective: To investigate the effects of T cell on the apoptosis and expression of apoptosis-related genes of RINm5f islet cells. Methods: Apoptotic cells were identified by PI method. The mRNA levels of bcl-2, bax, p53 and fas were evaluated by the method of semi-quantitative RT-PCR. Results: PHA, T cells or PHA combined with T cells caused time-dependent increase of apoptosis rate. In the process of apoptosis, PHA combined with T cells could induce significant increase of mRNA level of bax and fas. The expression of bcl-2 and p53 mRNA was decreased with no statistical significance. Conclusion:The T cell may initiate and exacerbate the rejection of islet transplantation. The changes of the ratio of inducible apoptosis to anti-apoptosis genes may be relevant to the mechanisms of apoptosis and fas and bax genes play an important role in the apoptosis process.
