摘要
目的:研究来氟米特对被动性Heymann肾炎(PHN)的治疗作用,为临床膜性肾病的治疗提供依据。方法:Wistar大鼠尾静脉注射抗FX1A血清,建立PHN模型;用BCA法测量24h尿蛋白变化,用光镜、电镜观察肾脏病理改变,并用RT-PCR观察nephrinmRNA表达改变。结果:成功复制了PHN模型来氟米特能减轻PHN大鼠蛋白尿,肾脏病变程度,抑制nephrinmRNA表达减少。结论:来氟米特对PHN有一定的治疗作用,可能是通过提高nephrin表达及减少免疫复合物沉积来实现这一作用。
Objective: To investigate the role of leflunomide in the treatment for PHN, and to provide evidence for membranous nephropathy in clinic. Methods: The rat PHN model was induced by injection of anti-FX1A serum through tail vein. Twenty-four hours urine protein, nephrin mRNA expression and glomerular lesions were detected by BCA, RT-PCR and light and electronic micro-scope, respectively. Results: The rat PHN model was establish-ed successfully. Leflunomide reduced urine protein, glomerular lesions of rat with PHN,and increased nephrin mRNA expression. Conclusion: Leflunomide may play a role in the treatment for PHN, possibly by increasing nephrin mRNA expression and decreasing immune complex deposits.
