摘要 目的改进高效液相色谱法测定人血清中普罗帕酮的方法.方法色谱柱:shim-pack vp-ODS柱(150 mm×4.6 mm,5 μm).流动相为甲醇:醋酸盐缓冲液:丁胺=60:39.5:0.5(v/v),醋酸滴定pH至4.7,流速1.0 ml/min.检测波长250nm.结果最低检出限为0.125 μg/ml.日内变异系数为5.1%~8.1%,日间变异系数为7.8%~9.0%.结论该法简便可行,值得在临床检验中推广应用. Objective A HPIE method has been established for the determination of propafenone in human serum.Methods The column was shim- pack vp- ODS(150 mm× 4.6 mm,5 μm). The mobile phase was methanol- acetate solution- butylamine (60:39.5:0.5, v/v), pH value was adjusted to 4.7 by acetic acid. The flow rate of mobile phase was 1.0 ml/min. The detection wavelength was 250 nm. Results LOD was 0.125μg/ml.The intra-day variance was 5.1%-8.1%, and the inter-day variance was 7.8%-9.0%.Conclusion This HPLC method is practicable in clinical lab oratory for determining propafenone concentration in serum.
