摘要
目的观察螺旋藻多糖及其与化疗药物阿霉素联合使用对MCF-7细胞增殖,端粒酶活性的影响,探讨藻类多糖的抗肿瘤机制。方法应用荧光免疫组化检测bcl-2基因,p53基因的表达;应用TRAP-ELISA法检测端粒酶活性;MTT法检测用药后的MCF-7细胞存活率。结果螺旋藻多糖和阿霉素联合给药可以显著降低bcl-2基因的表达,提高p53基因的表达,下调端粒酶活性。结论螺旋藻多糖联合阿霉素可能通过促进MCF-7细胞凋亡,下调端粒酶活性达到治疗乳腺癌的作用。
Objective To observe the effect of combination of polysaccharide from spirulina platensis (PSP) and chemotherapeutic drug adriamycin on MCF-7 cells proliferation and their telomerase activity to explore the anti-cancer mechanism of algae polysaccharide.. Methods The bcl-2 and p53 genes expression were detected by immunofluorescence methods, telomerase activity were determined by TRAP-ELISA, the survival rate of MCF-7 ceils after adding drugs was observed by MTT. Results PSP, adriamycin and combination all could decrease bcl-2 gene expression, increase p53 gene expression and down-regulate telomerase activity. Conclusions SPS combining with adriamycin may treat breast cancer probably via promoting MCF-7 cells apoptosis and down-regulating telomerase activity.
