颅内注射c-fos反义寡聚核苷酸对急性期脑出血周边组织的HSP70表达的影响

摘要 目的:研究c-fos基因对于急性期脑出血周边组织的影响及其作用机制。方法:采用在血肿中心微量注射、免疫组织化学法、原位末端标记的方法在大鼠脑出血模型上观察c-fos反义寡脱氧核苷酸(an tisense ob ligodeoxynucleotides,an ti-senseODN s)对HSP 70表达的影响。结果:an tisense ODN s能够抑制脑出血周边组织的c-fos阳性细胞表达(P<0.01),加重了血肿周边神经元的损伤。同时,HSP 70的表达也有下降的趋势(P<0.01),而且在c-fos表达的区域也有HSP 70的表达。结论:HSP 70基因的表达对急性期脑出血周边神经元起保护作用;HSP 70基因的表达可能部分通过c-fos调控。 Objective: To explore the possible role of c-fos involved in the neuronal protection in early intracerebral hemorrhage (ICH). Methods: Microinjection, immunohistochemistry and in situ end - labeling assay were applied to observe the effect of c-fos antisense obligodeoxynucleotides (ODNs) on the expression of heat shock protein 70(HSP70) in a ICH rat model. Results: Antisense ODNs inhibited the expression of c-fos positive cells (P 〈 0.01) , and aggravated injury around hematoma in ICH, and meanwhile, a descending tendency of HSP70 expression was observed in the same region after antisense ODNS treatment (P 〈 0.01). Furthermore, the distribution of c-fos expressed cells was parallel to that of ERCC1 positive ones. Conclusion: HSP70 may play a neuroprotective role and its expression may be partly modulated by c-fos expression and c-fos may protect the brain from ischemic damage.
