肥胖SD大鼠骨骼肌组织中SOCS-3mRNA的表达

摘要 目的:初步探讨肥胖SD大鼠骨骼肌组织中SOCS-3mRNA的含量与高leptin血症的关系。方法:用高脂饮食制备饮食诱导的肥胖(DIO)SD大鼠模型,应用放射免疫分析法检测血清leptin水平,应用RT-PCR半定量检测骨骼肌组织中SOCS-3mRNA的含量,并进行相关分析。结果:DIO大鼠存在高leptin血症,但其骨骼肌组织中SOCS-3mRNA的含量与对照大鼠没有差别,而且骨骼肌组织中SOCS-3mRNA的含量与大鼠的肥胖程度及血清leptin水平没有相关性。结论:DIO大鼠的leptin抵抗与骨骼肌组织中SOCS-3mRNA的含量无关。 Objective: To research whether there is a relationship between hyperleptinemia and SOCS-3 mRNA level in skeletal muscle in DIO rats. Methods: Weaning male SD rats were induced to be obese by diet rich in saturated fatty acids. Then the serum leptin and lipid levels were measured, at the same time the mRNA levels of SOCS-3 in skeletal muscle were measured by means of semi-quantitative RT-PCR technique. Results: The serum leptin level of DIO rats was higher than that of control rats significantly( 2.30±0.56 vs 0.98±0.51ng.ml, P〈0.001 ). There is no difference between SOCS-3 mRNA level in skeletal muscle of DIO rats and that of control rats (median were 0.83 and 0.80 respectively, P=0.432 ). When calculating the Spearman correlation coefficient, SOCS-3 mRNA level in rats' skeletal muscle did not correlate with obese degree of rats and serum leptin level( P〉0.05 ). Conclusion: It is not certain that SOCS-3 mRNA level in skeletal muscle was related to leptin resistance in DIO rats.
