摘要
目的观察小鼠在动情周期不同阶段腹腔注射(ip)内毒素(LPS)时卵巢一氧化氮(NO)和内皮素(ET)含量的变化及与血浆E2和P含量变化的关系。方法用阴道涂片法确定小鼠动情周期,分别用RIA法和分光光度法检于动情周期不同阶段ip LPS时卵巢NO、ET的变化及与E2和P变化的关系。结果卵巢NO含量分别于动情期(此时E2含量较高)、动情后期(此时P含量较高)及动情间期(此时E2和P含量均低)受LPS处理后均升高,但于动情间期的升高程度更加明显;卵巢ET含量分别于动情期和动情间期受LPS处理后升高,但于动情后期受LPS处理无明显变化。结论内毒素诱导的NO、ET的升高与动情周期有关,E2或/和P能通过部分或完全拮抗内毒素诱导的NO或/和ET的升高,从而对内毒素导致的机体损伤产生保护性作用。
Objective To investigate the correlation of estrous cycle and content of No and ET in the ovarian of LPS--treated mice. Methods The content of ET plasma E2 and P in the ovarian was determined with radio immunoassay (RIA)and the orarian content of No was determined with Spectrophotometer in mice of different stages of estrous cycle treated by lipopolysaccharide (LPS). Results The contents of No in ovaries of all group were significantly increased when LPS was given in estrus (with higher estradiol,E2), metestrus (with higher progesterone,P) and diestrus (with lower E2 and P) respectively, but was the highest when LPS was given in diestrus. The content of ET in ovaries was increased significantly when LPS was given both in estrus and in diestrus,but did not change remarkably when LPS was given in metestrus. Conclusion There is a correlation between the contents of .No and ET and estrous cycle. Estradiol and/or progesterone may have protective effect on endotoxin damage through partly or entirely inhibiting No and / or ET production.
