摘要
用限制性内切酶消化、提取鼠白细胞介素6cDNA全基因,并克隆至逆转录病毒穿梭质粒pZIP-neoSV(x)BamHⅠ位点,经斑点杂交,限制性内切酶消化,电泳鉴定,证实成功地构建了表达鼠IL-6反义RNA的逆转录病毒重组体,从而为研究白细胞介素6基因转移与治疗提供了物质基础。
The mouse IL-6 cDNA was isolated by restriction enzyme XhoⅠfrom plasmid TXM/ IL-6 cDNA and inserted into the BamH Ⅰsite of the retroviral vector pZIP to construct the recombinant plasmid pZIP IL-6.It was screened and identified by agrose gel electrophoresis, restriction enzyme Bgl Ⅱanalysis and nucleic acid hybridization, the results evidenced that pZIP IL-6 contained the correctly inserted IL-6 cDNA fragment. This study provides a fundament for using recombinant retroviru-mediated antisense IL-6 gene transfer and therapy.
