摘要
目的研究二磷酸果糖(FDP)对低温保存兔心肌组织蛋白质的影响。方法兔心肌组织分别在4℃保存于含5mmol.L-1FDP或无FDP的StThomas液中,提取心肌组织蛋白质经SDS凝胶电泳比较,肽质量指纹谱质谱分析和Westernblot鉴定蛋白质分子。结果肽质量指纹谱分析鉴定到β-肌球蛋白重链、莱恩素受体(ryanodinereceptor)、α肌动蛋白、电压依赖的阴离子通道蛋白1(VDAC1)和腺苷酸激酶3等5种蛋白质差异,Westernblot鉴定到β-肌球蛋白重链、蛋白激酶Cε(PKCε)和IgG重链3种蛋白质差异。加入FDP后心肌组织中β-肌球蛋白重链、α肌动蛋白和PKCε得到更好保存。结论FDP可以增加低温保存的兔心肌组织中一些与心功能相关的重要的蛋白质的保存量,以上所述的存在差异的蛋白质,在心脏低温保存条件研究中,具有作为生物标记分子的潜在应用价值。
Objective To study the effects of fructose diphosphate (FDP) on heart proteins of hypothermically preserved rabbit heart tissues. Methods Heart tissues were preserved under conditions with or without FDP in St Thomas solutions at 4℃. The proteins from heart tissues underwent the SDS-PAGE,Westem blot, and peptide mass fingerprint analyses.
Results Protein contents of beta-myosin heavy chain, alpha-actin, ryanodine receptor, VDAC1, PKCepsilon, and adenylate kinase 3 were different among control and experiment groups by peptide mass fingerprint analysis and Western blotting. Betamyosin heavy chain, alpha-actin, and PKCepsilon were better preserved after adding FDP. Conclusion In hypothermic preserved heart tissue, FDP addition in St Thomas solution increases the retained amounts of above mentioned proteins which are important for the normal heart function. Some of these proteins can be used as a biomarker for the study on hypothermic heart preservation.
