摘要 目的研究荭草苷对犬实验性心肌梗死的影响,以便为荭草苷的临床应用提供实验依据。方法采用麻醉开胸犬,通过结扎犬左冠状动脉前降支(LAD)产生急性心肌梗死模型,从心肌梗死面积、心肌梗死程度及血清酶学变化观察荭草苷的抗心肌缺血作用,并从自由基损伤等角度分析其可能的作用机制。结果荭草苷(0.5,1.0,2.0mg.kg-1)可降低血清肌酸磷酸激酶(CK)、乳酸脱氢酶(LDH)活性,并能提高超氧化物歧化酶(SOD)活性,显著降低心肌梗死面积。结论荭草苷对急性缺血心肌有明显保护作用,作用机制可能与对抗自由基损伤,增强体内抗氧化酶活性,减少心肌耗氧量,减少心肌梗死面积有关。 Objective To study the protective effects of orientin on experimental dog myocardial infarction. Methods Models of acute myocardial infarction were prepared by ligating the left anterior descending coronary artery in dog. The anti-myocardial ischemia action was assessed by determing the changes in myocardial infarction size ( MIS), degree of myocardial infarction and serokinases. Its probable mechanism of action was proposed in view of the resultant myocardial free radical injury. Results Orientin (0.5,1.0,2.0 mg·kg^-1 ) decreased the activities of serum creatine phosphokinase and lactic dehydrogenase ,and increased the activities of superoxide dismutase. At the same time ,it also decreased the MIS in dogs induced by coronary occlusion. Conclusion Orientin plays a prominent protective action on myocardial ischemia, probably by inhibiting oxygen free radical formation and enhancing endogenous antioxidase activity, reducing myocardial oxygen consumption and reducing MIS in dogs.
