昆虫全基因组DNA的保存及提取

Preservation and Extraction of Genomic DNA from Insects

以蚂蚁、天牛、蝗虫等昆虫为实验材料，建立了75％酒精常温浸泡密封保存和福尔马林常温浸泡密封保存昆虫样品2～4周后进行全基因组DNA提取的方法。基因组DNA经紫外分光光度法（260nm、280nm）、琼脂糖凝胶电泳、PCR检测，结果表明：从两种方法保存的样品中均能提取到纯净完整的基因组DNA，适用于分子生物学实验。

The method for extraction genomic DNA from insect sample, such as ants,longicorn beetles and grasshoppers,which stored in formalin or 75% ethanol for 2-4 weeks,is described. The results of 260 nm, 280 nm, ultraviolet absorption spectrophotometry,agarose electrophoresis and PCR detection showed that the genomic DNA, which was isolated from ants,longicorn and locust separately, is suitable for molecular biglogy experiments.
