用SSR标记建立玉米黄早四DNA标准指纹图谱的方法研究 被引量:11

摘要 DNA标准指纹图谱是分子检测技术用于玉米新品种选育及其DUS辅助评价的重要基础。通过玉米对黄早四DNA的SSR标准指纹图谱构建方法的研究。筛选出20对可以用于玉米DNA标准图谱构建的核心引物,它们的平均PIC值为0.8554,检测出的等位基因数是6~15个,平均8.55个。Bnlg125和Bnlg2162检测到了玉米黄早四的DNA特有谱带;8%的聚丙烯酰胺电泳体系可以将亲缘关系很近的玉米品种区分开来。对玉米DNA标准指纹图谱的建立方法和DNA取样方法进行了深入探讨。 The standard DNA fingerprint was an important tool in maize breeding and DUS testing of new variety. The method of building DNA standard fingerprint by SSR marker in Huangzao 4, a famous maize inbred line in China was studied. And 20 pairs core primers for DNA standard fingerprint building were screened out with PIC (polymorphism information content ) values of 0. 6754 - 0.903 by which 6 - 15 alleles were detected. And the characteristical bands of Huangzao 4 were detected by Bnlg125 and Bnlg2162. The cluster analysis with UPGMA from Nei's(1978) divided Huangzao 4 into 3 groups and 3 sub-groups. In addition, 8 % polyacrylarnide gel electrophoresis systems could detect more bands than 3 % metaphor gel electrophoresis, therefore it could be used for distinguishing maize varieties among close relative groups. The method for building DNA standard fingerprint of maize and DNA sampling were also discussed in this paper.
