摘要
目的观察通脉颗粒对动脉粥样硬化兔模型血浆VEGF水平及心肌侧支循环的影响。方法将40只日本大耳白兔随机分为空白对照组、模型对照组、通脉颗粒小剂量组、通脉颗粒大剂量组,每组10只,空白组普通饲料喂养,其余各组予高脂饮食(含5%猪油、0.5%胆固醇)喂养12周复制成动脉粥样硬化模型,其中后两组同时予大小剂量通脉颗粒灌胃,观察各组动物实验前后血浆VEGF水平变化、左室冠状动脉总数及狭窄冠状动脉百分比。结果各模型组动物血浆VEGF含量及冠脉数均明显增加,尤以大剂量组增加幅度为大,与其他各组比较差异均有显著性(P<0.05),且血浆VEGF含量与冠脉数呈明显直线正相关(相关系数0.906,P<0.05)。空白对照组冠状动脉正常,其余各组均有不同程度粥样斑块形成和冠脉狭窄,而狭窄冠状动脉百分比大小剂量组均比模型组小,且大剂量组最小(P<0.05)。结论通脉颗粒可延缓冠状动脉粥样斑块的发生发展,并通过升高血浆VEGF水平从而促进心肌侧支循环的形成,这可能是该药治疗冠心病心绞痛的重要机制之一。
[Objective] To observe the effects of TongMai granules on the level of plasma VEGF and heart collateral circulation in atherosclerosis rabbit model. [Methods] Forty Japanese big ear rabbits were randomly divided into negative control group, model control group, low dosage TongMai granules group and high dosage TongMai granules group, each group consisting of ten rabbits. The negative control group was fed on common diet, the other groups were high-lipid fed (5% pig fut, 0.5%cholesterol )to establish atherosclerosis model. The latter two groups were given low dosage and high dosage TongMai granules respectively. Plasma VEGF, the number of coronary arteries supplying left ventricle and the percentage of stenosis of coronary artery was observed in all the rabbits before and/or after the treatment. [Results] Plasma VEGF and the number of the coronary arteries in the model group increased signficantly, especially in the high dosage TongMai group, there was significantly difference compared with those of other groups. Plasma VEGF levels are correlated with the number of the coronary arteries (r =0.906, P 〈 0.05). Normal coronary arteries were seen in the negative control group while in the other groups atherosclerosis of various degrees and coronary atreries stenosis came into being.The percentage of stenosis of coronary arteries was lower in the TongMai group than those in the model group, with those of the high dosage group being the lowest (P 〈 0.05). [Conclusions] The mechanisms by which TongMai granules alleviates angina pectoris may be attributed to its properties of increasing the level of plasma VEGF, preventing and treating atherosclerosis and promoting collateral circulation formation.
