摘要
Moraxellasp.CY34能以丙酮醇为底物,以异丙胺为氨基供体,通过静息细胞转化法生产氨基丙醇。通过单因素实验对Moraxellasp.CY34的摇床产酶培养条件进行优化,得出较佳的培养基组合:甘油1%,蛋白胨1%,MgSO40.03%,NaCl 0.3%,CaCl20.01%,KH2PO40.2%,K2HPO40.2%,FeSO4.7H2O 0.001%,微量元素溶液0.1%,初始pH8.0。500 mL三角瓶的装液量以75 mL为宜。菌体培养36 h,菌体量、酶活均达到最高。经优化后酶活力达到4.67μmol/h.g cell。
Using acetol as the amino acceptor and isopropylamine as the amino donor, the synthesis of aminopropanol was conducted by the resting cell of Moraxella sp. CY34. The effect of single factor on the flask incubation of Moraxella sp. CY34 was investigated. The optimum medium for the yield of enzyme was included: glycerol 1%, peptone 1%, MgSO4 0.03%, NaCl 0.3%, CaCl2 0.01%, KH2PO4 0.2%, K2HPO4 0.2%, FeSO4·7H2 o 0.001%, with 0.1% solution containing trace dements, initial pH 8.0. The optimal broth volume was 75 mL/500 mL flask. Both biomass and enzyme activity could attain the maximum after incubation 36 h. The enzyme activity reached 4.67 μmol/h·g cell under the optimal condition.
