Expression of RAB5A and MMP-9 in colorectal cancer and their correlation

摘要 目的检测RAB5A、基质金属蛋白酶-9(MMP-9)在大肠癌组织中不同部位的表达及两者之间的相互关系。方法应用免疫组织化学SP法对46例大肠癌组织、距癌灶3cm的癌旁组织和23例非肿瘤患者大肠组织中RAB5A和MMP-9进行检测。结果46例大肠癌组织中,RAB5A蛋白的表达阳性率为100%(46/46),阳性表达位于胞质和胞膜;MMP-9的表达阳性率73.91%(34/46),阳性表达位于胞质。RAB5A和MMP-9表达在有淋巴结转移、Dukes分期晚及分化程度低的患者中表达程度高(P<0.05),MMP-9的表达还在大肠癌浸润度深的患者中表达程度高(P<0.05)。RAB5A和MMP-9的表达与性别、年龄、肿瘤部位均无关(P>0.05)。RAB5A和MMP-9的表达存在正相关(P<0.05,r=0.434)。结论RAB5A和MMP-9的表达有可能作为判断大肠癌恶性程度、转移的参考指标。 Objective To investigate the expression of RABSA and MMP-9 in colorectal cancer and their correlation. Methods By using immunohistochemical technique, the expression of RABSA and MMP-9 was detected in 46 colorectalcarcinomas, tumor-adjacent colorectal carcinoma tissues and23 normal tissues. Results In 46 cases of colorectal cancer, the positive expression rate of RABSA protein which was located in cytoplasm and on the membrane of the cells was 100% (46/46) and that of MMP- 9 which was located in cytoplasm 73.91% (34/46) respectively. The expression of RABSA and MMP-9 in colorectal cancer was high in the patients with lymph node metastasis, late Duke's stage and low differentiation (P〈0.05). The expression of MMP-9 in colorectal cancer was high in the patients with deep invasion (P〈0.05 ). The expression of RAB5A and MMP-9 had no relationship with gender, age and tumor site (P〉0.05). There was a positive correlation hetweenthe expression of RAB5A and MMP-9 (P〈0.05, r=0.434). Conclusion Expression of RAB5A and MMP-9 might be used a reference indicator for evaluating malignancy and metastasis of colorectal cancer.
