摘要
环状糊精葡萄糖基转移酶是催化淀粉水解为环状糊精的酶。以β-环状糊精葡萄糖基转移酶高产菌株LZ10为研究对象,分别对该菌发酵培养基的碳源、氮源和pH进行了单因子分析,并对该3个因素进行正交实验及验证,最终确定该菌的最佳发酵培养基配方:玉米粉2.5%,黄豆粉5%,NaCO30.2%,K2HPO4.3H2O0.13%,MgSO4.7H2O0.02%,pH为9.0。在该条件下,菌株LZ10产β-环状糊精葡萄糖基转移酶的酶活可达892.86u/mL酶液。
β-cyclodextrin glucanotransferase (β-CGTase) could catalyze starch and further transfer it into cyclodextrin (β-CD). A β-CGTase strain LZ10 was studied and the carbon source, nitrogen source and pH of its culture medium were analyzed. Through the orthogonal experiments of the three factors, the optimal culture conditions were concluded as follows: 2.5 % corn powder, 5 % soybean powder, 0.2 % NaCO3, 0.13 % K2HPO4·3H2O, 0.02 % MgSO4·7H2O and pH as 9.0. Under such conditions, the highest β-CGTase activity could achieve 892.86 Unit per micro-liter fermentation liquid.
