摘要
在pH4.0的三羟甲基氨基甲烷-HCl缓冲溶液中,结晶紫与蛋白质结合形成稳定的深红色复合物。该复合物λmax为575nm,其表观摩尔吸光系数为3.25×105L/mol.cm,蛋白质含量在0.0~80.0μg/mL范围内服从朗伯比尔定律,检出限为6.47μg/mL,回收率为97.0%~102%,此法可用于黄酒中总蛋白质含量的测定。
Crystal violet reacted with protein to form stable scarlet compounds in Tfis-HCl buffer solution (pH value as 4). The maximum absorption of the compounds was 575 nm, its molor absorptivity was 3.25×10^5L/mol^-1·cm^-1, and Beefs law was obeyed for protein in the rang of 0.0-80.0 μg/mL and the detection limit and the recovery were 6.47 μg/mL and 97 %- 102 % respectively. This method could be used in the determination of total protein content in yellow rice wine.
