人类肾癌中AKR1C2基因的表达及其临床病理意义 被引量:1

摘要 目的:为了探讨人类肾癌中AKR 1C 2基因的表达特征及其临床病理意义。方法:选择30例肾癌手术切除患者的肿瘤标本,利用RT-PCR、免疫组织化学染色检测肾癌及相应的癌旁组织中的AKR 1C 2基因的表达,同时分析肿瘤中AKR 1C 2表达与患者临床病理特征间的相关性。结果:RT-PCR结果显示,30例肾癌组织中AKR 1C 2 mRNA均为上调表达,其中6例癌旁组织中未见AKR 1C 2 mRNA的表达。免疫组织化学结果显示,30例肾癌组织中AKR 1C 2蛋白均为上调表达,其中13例癌旁组织中未见AKR 1C 2蛋白的表达。AKR 1C 2的表达与肾癌患者临床病理特征间的相关性分析显示,AKR 1C 2基因的L I和E I与肿瘤患者的R obson分期、肿瘤分化、肿瘤病理类型及其转移呈显著正相关。结论:提示人类肾癌组织中AKR 1C 2在转录与翻译水平上均呈过表达,可能有助于肾癌的发生发展。 Objective : To investigate the expression of AKR1C2 gene in human renal carcinoma and its clinicopathological significance . Methods: The expression of AKR1C2 gene was detected by RT- PCR and Immunohistochemistry (IHC ) and its relationship with patient's clinico-pathological characteristics was analysed among 30 cases of renal carcinoma tissues and their adjacent noncancerous tissues. Results: The RT-PCR and Immunohistochemistry(IHC)results both showed that the expression of AKR1C2 was up-regulated in all 30 cases of renal carcinoma tissue and its expression was not detected in adjacent noncancerous tissues from 6 , 13 out of 30 cases respectively. The analysis of relationship between expression of AKR1C2 and clinico-pathological characteristic among these patients showed that the label index (LI)and the expression index (EI) of AKR1C2 gene were positively related to the renal carcinoma's Robson stage, tumor differentiation grade, the tumor pathological type and metastasis. Conclusion: It was suggested that AKR1C2 gene was over expressed in human renal carcinoma tissue on the level of transcription and translation and it may contribute to the renal carcinogenesis and its development.
