摘要
人乳头瘤病毒E5是一种转化作用的癌蛋白,是细胞膜或内膜整合蛋白。人乳头瘤病毒E5在感染的细胞中表达,主要在感染细胞克隆早期的繁殖、扩张中起重要作用。它干预生长因子受体,干扰周期蛋白和周期蛋白激酶,促进病毒癌基因转化,抑制抑癌基因表达,激活启动子促进病毒繁殖,并通过多种机制促使损伤细胞,通过细胞周期,使宿主细胞增殖、分化延缓、恶性化。近年的研究揭示了不同类型人乳头瘤病毒癌蛋白的功能差异,为人乳头瘤病毒诱导细胞永生化和致癌分子机制的研究提供了依据。该文将从E5对信号转导途径的影响,对细胞周期的影响,与癌基因和抑癌基因的相互作用,反式激活作用和变异几方面综述E5的研究进展。
HPV E5 is a kind of transforming cancer albumen, which intima. E5 is expressed in cells infected by HPV, which mainly exists belongs to integrating protein on cell membrane and tunica in Golgi apparatus, endosarc net velum and karyotheca,and plays important roles in reproduction and expansion of early infected cells clone. It can intervene growth factor receptor, disturb cycle protein and kinase, promote transformation of virus gene to oncogene,inhibit expression of cancer suppression gene, activate promoter to initiate reproduction of the virus and through many mechanisms it damage cells and through cell cycles it induces multiplication of host cells and defers differentiation and induces deteriorism. Some researches in recent years revealed the function difference in different HPV cancer protein,which provide evidence for the research on cell permanence and molecular mechanism of carcinogenesis. In this paper,we summarized the evolution in research on effects of E5 on the approach of signal conduction, and cycles of the cell,interactions with oncogene and cance suppressing gene, trans-form activation and variation.
