慢性应激大鼠抑郁模型的建立及其评价

Establishment and Assessment of Rat Model of Chronic Unpredictable Mild Stress Depression

目的探讨建立慢性应激抑郁大鼠模型的实验方法及模型的有效性。方法利用慢性轻度不可预见性应激(CUMS)制作抑郁模型,观察动物的体重和摄食量的变化,用敞箱试验及液体消耗实验测定动物行为。结果大鼠抑郁模型在第27天建立成功。抑郁组大鼠敞箱试验得分、液体消耗试验中糖水消耗和糖水偏爱百分比均明显下降。结论慢性轻度不可预见性应激可以成功建立大鼠抑郁模型。

Objective To explore the validity and reliability of experimental rat model of chronic unpredictable depression. Methods The rat model of depression was established by exposing to an unpredictable sequence of stressors. The changes in body weight and food intake in the animals were observed. By open--field test and fluid consumption test,the behaviors of the animals were determined. Resuits The rat model of depression had been established successfully at 27th day. In the depressive group, the rats showed reduced squares crossing and rearing in open-field test, a significandy reduced consumption of preference for sucrose solutions and an increased pure water consumption as compared with con- trol group,but the total water consumption among all the experiment groups had no statistic significance. Conclusions The rat model of depression can be established successfully by exposing to an unpredictable sequence of stressors.
