柱前衍生化法测定血浆中醋酸精氨酸的浓度

摘要 目的:建立柱前衍生化高效液相色谱法测定血浆样品中醋酸精氨酸浓度的方法。方法:衍生剂为2,4-二硝基氟苯,色谱柱为氨基酸分析专用ODS色谱柱,流动相为50%乙腈-0.1mol/L醋酸钠溶液(29:71),流速为1.2ml/min,检测波长为360nm,柱温为30℃。结果:醋酸精氨酸检测浓度线性范围为10~160μg/ml(r=0.9998),高、中、低3种浓度血浆样品的回收率分别为(96.64±9.69)%、(90.94±4.70)%、(92.72±4.48)%,精密度〈3.91%。含药血浆在冰冻条件(-24℃)下至少可以稳定15d。结论:本法简便可行,适用于醋酸精氨酸的血药浓度监测及其药动学研究。 OBJECTIVE: To establish a pre - column derivatization HPLC method for the determination of arginine acetate in plasma sample. METHODS: 2,4-dinitro-1-fluorobenzene (DNFB) was taken as derivative, the chromatographic column was ODS column, which was used specially for amino acids analysis, the mobile phase was composed of 50% acetonitrile 0. 1mol/L sodium acetate solution(29 : 71), the flow rate was 1.2ml/min,the detection wavelength was 360nm and the column temperature was 30℃ . RESULTS: The linear concentration range of arginine acetate was 10-160μg/ ml( r = 0.9 998), the respective recovery rate of high, medium and low concentration of plasma samples was(96.64 ± 9.69) %, ( 90.94 ±4.70) % and (92.72 ± 4.48)%, with RSD less than 3.91%. The drug containing plasma was stable at least in 15 days under freezing condition(-24℃ ) . CONCLUSIONS: The elaborated method is simple and practicable, and suitable for monitoring of blood drug level of arginine acetate and it's pharmacokinetic study.
