高效液相色谱法测定万应胶囊中胡黄连苷Ⅰ和胡黄连苷Ⅱ的含量

Content Determination of Picroside Ⅰ and Picroside Ⅱ in Wanying Capsule by HPLC

目的：建立以高效液相色谱法测定万应胶囊中胡黄连苷Ⅰ和胡黄连苷Ⅱ含量的方法。方法：色谱柱为Diamonsid^TM C18，流动相为乙腈-水-冰醋酸（20：80：0．5），检测波长为266nm，流速为1．0ml／min。结果：胡黄连苷Ⅰ、胡黄连苷Ⅱ检测浓度分别在9．55～95．5μg／ml（r=0．9997）、15．6～156μg／ml（r=0．9998）范围内线性关系良好，平均回收率分别为99．7％（RSD=2．46％）、101．2％（RSD=2．35％）。结论：本方法简便、快速、准确、回收率好，可用于该制剂的质量控制。

OBJECTIVE：To determine the content of picroside Ⅰ and picroside Ⅱ in wanying capsule by HPLC.METHODS： The content determination was conducted with Diamonsil- C18 as chromatographic column under room temperature, the mobile phase consisted of acetonitrile- water-glacial acetic acid（20 ： 80 ： 0.5）, the detection wavelength was 266 nm and the flow rate was 1.0ml/ min.RESULTS： Good linear relations were achieved when the concentration ranges of picroside Ⅰ and picroside Ⅱ were 9.55～95.5μg/ml（r = 0.9 997） and 15.6～ 156μg/ml（r = 0.9 998）, respectively, the average recoveries for picroside Ⅰ and picroside Ⅱ were 99.7% （RSD = 2.46%） and 101.2% （RSD = 2.35%）, respectively.CONCLUSIONS： The method is simple,fast and accurate,and in which high recovery rate was achieved,thus it can be used for the quality control of the preparation.
