硫酸镁对脑损伤大鼠神经细胞凋亡及Caspase-3表达的影响

摘要 目的研究硫酸镁对创伤性脑损伤大鼠神经细胞凋亡及Caspase-3基因表达的影响。方法Wistar大鼠48只,随机分为6组,每组8只。采用Feeney法造成TBI模型。损伤组大鼠在致伤后5min内分别按不同剂量给予25%MgSO4腹腔注射。24h后检测细胞凋亡及Caspase-3表达情况。结果颅脑创伤后大鼠脑组织中神经细胞凋亡及Caspase-3基因表达均显著增加,使用MgSO4治疗后神经细胞凋亡显著减少,Caspase-3基因表达降低。不同剂量MgSO4组之间神经细胞凋亡与Caspase-3基因表达未见差异显著性。结论硫酸镁可显著抑制创伤性颅脑损伤大鼠神经细胞凋亡及Caspase-3基因活化,具有显著的脑保护作用。 Objective To study the effects of magnesium on apoptosis and the expression of caspase - 3 gene in TBI rats. Methods 48 SD rats were randomly divided into 6 group each consisted of eight, cortical contusion in rats was induced with the Feeney model. Five minutes after TBI, the rats were treated with different magnesium dose. The results of apoptosis and the expression of caspase - 3 gene in TB1 rats were determined 24 hours after injection of magnesium sulfate. Results The apoptosis and expression of caspase - 3 gene in TBI rats increased significantly. The apoptosis and expression of caspase - 3 gene decreased remarkably after treatment with magnesium sulfate. There are no different between the groups treatment with different dose of magnesium sulfate. Conclusion Magnesium sulfate can significantly restrain the apoptosis and the expression of caspase - 3 gene, which might have clinical value for treating brain injury rats.
