摘要
目的探讨选择性环氧合酶-2(Cox-2)抑制剂Celebrex对体外培养的人胆囊癌GBC-SD细胞系增殖的影响。方法采用MTT比色法检测Celebrex对细胞系生长的影响;TUNEL染色法观察细胞凋亡指数,并用流式细胞仪定量分析;透射电镜和荧光显微镜检测细胞凋亡。结果Celebrex抑制GBC-SD细胞系的生长呈剂量依赖性;4 0,8 0,1 2 0,1 6 0μmol/L浓度的Celebrex对GBC-SD细胞的生长抑制率分别是1 8.7 7%,2 5.3 2%,4 6.5 8%和5 2.1 9%(P<0.0 1)。流式细胞仪定量分析在4 0,8 0,1 2 0,1 6 0μmol/L浓度下的细胞凋亡率分别为(8.5 1±1.4 4)%,(1 2.4 0±0.8 7)%,(2 6.3 7±1.7 2)%和(4 3.2 1±0.3 9)%,与对照组(4.8 7±0.5 5)%比较有显著的统计学差异;各组间两两比较差异亦有显著性(均P<0.0 1)。透射电镜和荧光显微镜下能观察到胞核浓缩、碎裂以及凋亡小体的形成。结论Celebrex可以有效地抑制人胆囊癌GBC-SD细胞系的增殖并诱导其凋亡。
Objective To investigate the effect of cyclooxygenase-2 inhibitor Celebrex on the growth of gallbladder carcinoma GBC-SD cell line. Methods Cell growth suppression was counted by MTr method. Apoptotic index (AI) was evaluated by TUNEL staining. The apoptotic rate was counted by flow cytometry ( FCM ) , fluorescence microscopy ( FM ) and transmission electron microscopy ( TEM ) . Results Celebrex inhibiting the growth of GBC-SD cell line was dose-depend. The growth inhibition rate with 40μmol/L, 80μmol/L, 120μmol/L and 160μmol/L was 18. 77%, 25. 32%, 46. 58% and 52. 19% respectively, ( P 〈 0.01 ) . Apoptotic rate of GBC - SD cell with 40 μmol/L, 80 μmol/L, 120 μmol/L and 160μmol/L Celebrex was (8.51 ± 1.44)% ,(12.40 ±0.87)% ,(26.37 ±1.72)% and (43.21 ±0.39 ) % respectively, and was significant difference in relation to the control group ( 4.87 ±0.55 ) % . Apoptotic bodies and cell shrinkage and fragmentation was observed by FM and TEM. Conclusions Celebrex can effectively inhibit the growth of gallbladder carcinoma GBC-SD cell line, and induce the apoptosis.
