任脉电针对脑缺血大鼠侧脑室下区神经干细胞增殖与分化的影响 被引量:28

摘要 目的观察任脉电针对脑缺血大鼠侧脑室下区神经干细胞增殖与分化的影响。方法制备大脑中动脉闭塞模型(MCAO),两小时后再灌注。实验大鼠随机分为手术对照组和任脉电针组,分别于造模成功7天、14天和28天后观察侧脑室下区神经干细胞增殖与分化的变化。结果与手术对照7天组(C-7d)相比,任脉电针7天组(R-7d)的5-溴脱氧尿苷(Brdu)阳性细胞、5-溴脱氧尿苷/胶质纤维酸性蛋白双标细胞(Brdu/GFAP)、5-溴脱氧尿苷/巢蛋白双标细胞(Brdu/Nestin)、5-溴脱氧尿苷/神经元特异性烯醇化酶双标细胞(Brdu/Nse)数量差异无显著性(P>0.05);而在14天组及28天组中,任脉电针14天组(R-14d)、任脉电针28天组(R-28d)Brdu阳性细胞及Brdu/Nestin、Brdu/Nse双标细胞数量分别较手术对照14天组(C-14d)、手术对照28天组(C-28d)差异有显著性(P<0.05)。在28天组,R-28d的Brdu/GFAP双标细胞明显多于C-28d(P<0.01)。结论任脉电针能促进脑缺血大鼠侧脑室下区神经干细胞的增殖与分化,适当促进增殖的神经干细胞向星形胶质细胞方向分化。 ObJective: To observe effects of electroacupuncture (EA) at The Conception Vessel on proliferation and differentiation of nerve stem cells in the subventricular zone of the lateral ventricle of cerebral ischemia rats. Methods:The middle cerebral artery was blocked for 2 hours and then reperfused. Model rats were randomly divided into two groups :control group and EA group. Changes of differentiation and proliferation of nerve stem cells were observed at 7, 14, 28 days after successfully modeling. Results: Compared with the control group, there was no significant difference (P~ 0. 05) in the number of 5-bromodeoxyuridine (Brdu) positive cells, Brdu/GFAP, Brdu/Nestin and Brdu/Nse double-labeled cells in the subventricular zone in EA group 7 days after modeling. However,in the 14-day group and the 28-day group,the number of Brdu positive cells and Brdu/GFAP ,Brdu/Nestin, Brdu/Nse double-labeled cells significantly increased as compared with the control group(P〈0. 05 or P〈0. 01 ). Conclusion :EA at The Conception Vessel promotes differentiation and proliferation of nerve stem cells in the subventricular zone of the lateral ventricle of cerebral ischemia rats,and may promote proliferation of the nerve stem cell towards astrocytes.
