摘要 [目的]了解CCl4对小鼠机体的影响,建立肝损伤动物模型,用于其保肝功能保健食品筛选。[方法]小鼠按0.01 ml/kg.BW一次性经口灌胃染毒,设对照组及3个浓度(25、35和40 mg/kg.BW),每个浓度按24、48和72 h采血,测定血清ALT、AST、TG和GLU,计算肝体比并进行常规组织病理学检查。[结果]25 mg/kg.BW CCl4染毒24 h能升高小鼠血清AST值,肝重增加,但肝体比与对照组类似(P>0.05);3个浓度CCl4染毒24 h对小鼠血清TG值均无影响(P>0.05);35及40 mg/kg.BW CCl4在各时间段均能提高血清ALT、AST、TG值及肝体比,增加病变总积分,降低GLU值(P<0.01)。[结论]35 mg/kg.BW CCl4对小鼠一次性经口染毒24 h,是较理想的检测其保肝功能保健食品的动物模型。 [Objective] To study the effect of carbon tetrachloride (CCl4) on mice liver damage and setup an animal model for the screening of liver-protecting healthy food. [Methods] Mice were orally given CCl4 0.01 ml/kg. BW once and the blood samples were collected in 3 groups with concentrations (25, 35, 40 mg/kg BW) and one control group in different periods (24 h, 48 h and 72 h). ALT, AST, TG and GLU of serum were detected and liver index were examined with routine histopathology. [Results] After exposure to 25 mg/kg. BW CC14 for 24 h, serum AST and liver weight increased, but liver index was similar to control group (P 〉0.05). Serum TG levels in 24 h were similar (P 〉0. 05). Serum ALT, AST, TG and liver index increased by 35 mg/kg. BW and 40 mg/kg. BW in 24 h, 48 h and 72 h. The scores of pathological changes increased and GLU decreased (P 〈0. 01). [Conclusion] An ideal mice animal model for evaluating healthy food for liver protection can be set up by orally CCl4 once with 35 mg/kg. BW for 24 h.
