TPA诱导外周血单个核细胞聚集的研究

A Study on TPA Induced Peripheral Blood Mononuclear Calls Aggregation in Normal Subjects and Acute Leukemia

应用不同浓度TPA诱发聚集,光电比色法检测35名正常人、43例白血病患者外周血单个核细胞(PBMC)以及K562和L1210细胞聚集率。5～1000ng/mlTPA可诱导PBMC稳定聚集,但不能使K562白血病细胞株细胞发生聚集。当K562和L1210细胞与10ng/mlTPA共同孵育5天后,其将对TPA出现低于正常PBMC聚集反应。TPA诱导的白血病患者PBMC聚集率明显低于正常人,且与幼稚细胞数呈负相关。

Using different concentrations (5ng/ml^2000ng/ml) of TPA as a aggregating agent, with colorimetric analysis,we studed PAg in 35 normal subjects and 43 acute leukemia patients as well as K562 and L1210 cell line cells. The result showed that TPA induced PAg of normal subjects was quite stable. At the concentration from 5ng/ml to 1000ng/ml, with the increasing of TPA concentration, the rates of the PAg was not changed(P>0. 05). when the concentration of TPA was over l000ng/ml, 1 minute (PAgl' )and maximum PAg(PAgM)was still the same,but the 5 minute PAg(PAg5' )was obviously decreased due to the disaggregation. TPA can not induce K562 and L1210 cell to aggregate. when the K562 or L1210 cell was mixed with the normal PBMC in differant rates and the PAg of this mixed cells was detected,we found that the PAgM rate was high negative related with the rates of K562 or L1210/normal PBMC(r = ?. 96 and ?0. 93). when the K562 cell was pretreated with 10ng/ml TPA at the condition of 37℃5%CO2 for 5 days, then washed and induced with 10ng TPA,aggregation was appeared and the PAgM was 17. 14±5. 88%, much lower than that of normal PBMC (65. 23±9. 53% ,P<0. 01). TPA induced PAg rate of PBMC from 43 leukamia patients was 21. 30±12. 71 ,destinctly lower than that of normal PBMC too,and the rate of PAg was negative raleted with percentage of leukemia cells(r= ?. 78). The results indicated that, unlike normal PBMC, leukemia cells lack a adhesive factor which can react to TPA to make the cells aggregate. When leukemia cells were pretreated with TPA for certain time,they can be stimulated by TPA to express this factor.
