摘要
目的:研究survivin在不同血液肿瘤细胞株(Jurkat,Raji,EL-4,K562,K562/AO2)的表达,并探讨其与相关因素的关系。方法:应用RT-PCR技术检测survivin mRNA在各细胞株的表达,用免疫组化法检测survivin蛋白的表达,流式细胞仪检测细胞周期。结果:survivin mRNA在4种人源性细胞株(Jurkat,Raji,K562,K562/AO2)中均有表达,耐药株K562/AO2高于其他细胞株;免疫组化结果显示survivin蛋白在胞核与胞浆均有表达,且表达水平与mRNA水平一致;细胞周期结果显示,survivin表达较高的细胞株G2/M期细胞比例和增殖指数较高。结论:survivin基因在血液肿瘤细胞株中普遍表达,survivin高表达可能与肿瘤细胞增殖及耐药有关。
Objective: To explore the expression of survivin in different haematological tumor cell lines (Jurkat, Raji, FL-4, K562, K562/AO2). Methods: The levels of survivin mRNA and survivin protein were detected by RT-PCR and immunohistochemical staining, respectively, and the cell cycle profile was measured by flow cytometry. Results: Survivin mRNA expressed in four human cell lines (Jurkat, Raji, K562, K562/AO2) with varying levels and the roost highly in the K562/AO2. Survivin protein was detected in both nucleolus and cytoplasm in the four human cell lines, whose level is consistent with that of survivin mRNA. The higher the survivin mRNA expression was, the higher the proportion of G2/M phase cell and the proliferation index were. Conclusion: Survivin gene expresses in haematological tumor cell lines at large, its high expression may relate with tumor cell proliferation and drug resistance.
