消除Ia阳性细胞及短程应用CyA诱导大鼠甲状旁腺同种异体移植物长期存活 被引量:1

摘要 本实验采用单抗加补体方法消除大鼠甲状旁腺(PTG)移植物中的Ia阳性细胞,处理后移植物分泌功能保存良好。将其移植到同种受体肾包膜下,移植物平均存活时间达60d,与未处理移植组(14d)相比,P<0.01。若受鼠接受移植前应用环孢素(CyA)3d(2.5mg·kg^(-1)/d),则可使60%经处理的PTG同种异体移植物存活期大于150d;与未处理短程应用CyA组(70d)相比,P<0.01。说明消除Ia抗原阳性细胞与术前短程给予受体CyA联合应用,可使大鼠PTG同种异体移植物获得长期存活。 In this study,Ia ̄+ donor cells of rat parathyroid allografts were depleted by anti-Ia monoclonal antibody plus complement.The secreting function of treated glands as well preserved.Then we transplanted the treated glands into subcapsular of the recipient kindey,and the median survival time(MST) of the allografts were 60 days, compared with 14 daysin the fresh PTG group(P<0.01 ).If the recipients received 25 mg/kg cyclosponine forthree days before transplantation,the MST of about 60%PTG allografts was more than 150days,with P<0.01 in comparison with the control group(short course CyA alone,MST:70days ). These results indicate that long-term survival of rat parathyroid allografts can beachived by depletion of la ̄+ donor cells and short course pretreatment of the recipient withCyA。
