双歧杆菌完整肽聚糖对重组乙肝表面抗原的佐剂效应 被引量:4

摘要 目的探讨双歧杆菌完整肽聚糖对重组乙肝表面抗原的吸附作用及其佐剂效应。方法从双歧杆菌细胞壁提取完整肽聚糖,以不同方式与重组乙肝表面抗原结合,检测其对重组乙肝表面抗原的吸附作用;制备免疫原接种BALB/c小鼠,观察小鼠免疫后状态和生长情况,检测小鼠血清特异性抗体应答情况的变化。结果在磷酸盐缓冲液中,完整肽聚糖对重组乙肝表面抗原的吸附呈剂量依赖性,作为佐剂免疫小鼠后无毒副反应出现,小鼠血清抗体阳性率和抗体滴度明显提高,IgG2a/IgG1增大。结论双歧杆菌完整肽聚糖能够吸附重组乙肝表面抗原,增强机体相应的体液免疫应答和细胞免疫应答,具备良好的免疫佐剂效应。 Objective To investigate the adjuvant effect of bifidobacterial whole peptidoglycan (WPG) on recombinant hepatitis B surface antigen. Methyls Extracting WPG from bifidobacterial cell wall and detecting it's physical-chemical characteristic. The adsorption of WPG on recombinant hepatitis B surface antigen were checked and evaluated through ELISA. Recombinant hepatitis B surface antigen in combination with bifidobacterial WPG was inoculated into BALB/c mice, post-immunization state and condition of growth were observed, and specific antibody level in serum was detected. Results Adsorption of the intact molecules of WPG on recombinant hepatitis B surface antigen was a dose-dependent process. No adverse effect was observed, positive ratio and titer of specific antibody in serum were increased markedly as well as the ratio of IgG 2 a to IgG 1 . Conclusion Bifidobacterial WPG can bind to recombinant hepatitis B surface antigen, and promot humoral immunization as well as cellular immunization simultaneously indicating a perfect adjuvant potential.
