感耦等离子体质谱法和发射光谱法测定卡铂-乳酸/羟基乙酸共聚物微球中卡铂含量的研究 被引量:6

摘要 目的:建立感耦等离子体质谱和感耦等离子体发射光谱直接测定卡铂-乳酸/羟基乙酸共聚物微球中卡铂含量的方法,并比较不同的样品前处理方法对测定结果的影响。方法:采用不同方法消化卡铂-乳酸/羟基乙酸共聚物微球,分别采用感耦等离子体质谱法和感耦等离子体发射光谱法测定,考察方法的线性、精密度和回收率。结果:微波消化法能够完全消解卡铂-乳酸/羟基乙酸共聚物微球;ICP-AES 法在0.5~10μg·mL^(-1)范围内线性良好,测定标准曲线为 Y=1003.959C+4.126,r=0.9999;仪器进样精密度2.10%,方法精密度为2.10%;平均加标回收率为97.5%±5.72%。ICP-MS 法0.2~20ng·mL^(-1)范围内线性良好,测定标准曲线为 Y=0.1254C+0.0042,r=0.9999;仪器进样精密度0.64%,方法精密度为1.56%;平均加标回收率为102.9%±4.29%。结论:采用微波消化样品的方法,ICP-AES 法和 ICP-MS 法均能比较准确地测定样品中铂含量,但两种方法适用的浓度范围不同,应根据样品浓度选择相应的方法进行含量测定。 Objective:To establish a direct determination method for carboplatin/poly(lactic -co -glycolic acid) (PLGA) microspheres by ICP - MS and ICP - AES. The influence of sample treatment on determination was also studied. Methods:Carboplatin/PLGA microspheres were pretreated by microwave digestion or heating plate -acid digestion and then determined by ICP - MS and ICP - AES, respectively. The linearity, precision and recovery of the determination methods were studied. Results: Carboplatin/PLGA microspheres can be completely digested by microwave digestion. In the range of 0. 5 - 10 μg·mL^-1 platinum ( Pt), ICP - AES gave a determination regression curve Y= 1003. 959C +4. 126 with r =0. 9999 ;Pt concentration was as low as 0. 2 -20 ng·mL^-1. Can be well determined by ICP - MS The determination regression curve was Y = 0. 1254C + 0. 0042, r = 0. 9999. The sampling precision of ICP - AES and ICP - MS was 2. 10% and 0. 64% ,respectively. The determination precision of the two methods was 2. 10% and 1.56% ,respectively,with the average recovery being 97. 5% ±5.72% and 102. 9% ± 4. 29%, respectively. Conclusions :When sample were preterated by microwave digestion, carboplatin/PLGA microspheres can be accurately determined by both ICP - AES and ICP - MS. The two methods had different sensitivity and should be chosen according to sample concentration.
