摘要
目的:建立同时测定硫酸软骨素主成分含量和掺入的盐酸氨基葡萄糖含量的高效液相色谱法。方法:采用 C_(18)色谱柱(用极性小分子封尾),以乙腈-10mmol·L^(-1)戊烷磺酸钠溶液(10:90)为流动相,流速为0.8mL·min^(-1),检测波长195nm。结果:以响应值对浓度作图,硫酸软骨素在0.01~0.75mg·mL^(-1)内呈很好线性,r=0.9996,盐酸氨基葡萄糖在0.01~1.50mg·mL^(-1)内呈很好线性,r=0.9994;硫酸软骨素含量测定的 RSD=0.67%,盐酸氨基葡萄糖含量测定的 RSD=1.26%(n=12)。结论:该方法重复性、再现性好,适合软骨素产品的定量测定。
Objective:To establish a RP - HPLC method for simultaneous determination of Chondroitin Sulfate and Glucosamine Hydrocbloride. Methods: Using C 18 (embedded polar group)column. The mobile phase was Acetonitrile - 10 mmol ·L^-1 Sodium 1 - pentanesulfonate ( 10: 90). The flow rate was 0. 8 mL·min^-1. The detection wavelength was 195 nm. Results:The calibration curves of Chondroitin Sulfate were liner in the range of 0. 01 - 0. 75 mg ·mL^-1 (r =0. 9996). The calibration curves of Glucosamine Hydrocbloride were liner in the range of 0. 01 - 1.50 mg·mL^-1(r =0. 9994). The contents of determination of Chondroitin Sulfate, RSD =0. 46% ,The contents of determination of Glucosamine Hydrochloride, RSD = 1.25 % (n = 12 ). Conclusion:The method is accuracy, convenience and good repeatability.
