Study on cultivation conditions for aflatoxin producing by aspergillus flavus AS3.4408 using Box-Behnken design

摘要 在Plackett-Burman试验设计结果基础上,采用Box-Behnken设计实验、响应曲面法(Responsesurfacemethodology,RSM)分析,对影响黄曲霉AS3.4408最适产黄曲霉毒素的关键培养条件温度、装液量和培养时间进行了探讨。结果表明,在培养温度为27~29℃、装液量为55~65mL、培养时间为7~10d条件下,每毫升发酵液可获得>500ng黄曲霉毒素;通过对毒素曲面方程和二次多项回归方程解逆矩阵得知在培养温度、装液量和时间分别为27℃、64.5mL和9d时,黄曲霉AS3.4408产黄曲霉毒素的最大预测值为606ng/mL发酵液,验证试验证实了该方程的预测值与实际值之间具有较好的拟合度。 Response surface methodology (RSM) based on the Box-Behnken design of experiment was used to optimize the aflatoxin production by aspergillus flavus AS3.4408. The critical factors selected for the investigation were temperature of cultivation, volume and time based on the results of previous Plackett Burman design. By analyzing the response surface plots, the optimum ranges of cultivation temperature, medium volume and time for obtaining over 500ng of aflatoxin are in 27-29℃, 55 -65mL and 7-10d, respectively. By solving the inverse matrix from the quadratic regression equations, the optimal conditions for obtaining 606ng/mL of aflatoxin are 27%, 64.5mL and 9d. The experimental data under various conditions have validated the theoretical values.
