芹菜总黄酮的提取及含量测定 被引量:20

摘要 研究甲醇溶剂提取芹菜总黄酮的提取效果和光度法测定总黄酮的实验条件,并使用正交实验确定了最佳提取条件。结果表明甲醇最佳提取条件为A3B2C3,即以90mL的80%甲醇回流2h。光度法在2~100μg/mL测定范围内符合比耳定律,其回归方程A=0.025+0.035C(μg/mL),线性关系良好,变异系数为2.98%~5.48%,回收率为96.8%~102.6%。最低检测限为1μg/mL。测定方法简单快速,具有一定的测试准确度和灵敏度,可用于实际样品中黄酮类化合物的分析。 The optimum extracting condition by methanol and the determination method by spectrophotometric method were studied. The results show that the optimum extracting condition is A3B2C3, that's to use 90mL 80% methanol to extract with backwatedng system for 2h. Within in the concentration range 2-100gg/mL of flavones was well linear with the absorption when using the spectrophotometric method, and the linear equation is A=0.025+0.035C (μg/mL), RSD and the recovery are 2.98%-5.48% and 96.8%-102.6%, respectively. The determination limit is 1μg/mL, this method is simple, quick and accurate, could be used for the determination of flavones in real samples.
