摘要
采用正交试验方法,对芦笋中芦丁的最佳提取工艺进行了研究,以反相高效液相色谱(采用C18反相色谱柱,以甲醇∶水∶磷酸(60∶40∶0.5)为流动相,流速为1mL/min,检测波长为254nm)等方法对提取的芦丁进行了定性、定量鉴定。研究结果表明,在75℃条件下用25倍的75%乙醇浸提5h,提取效果最好,浸提率为78.7%,测定方法的平均回收率为97.83%,测定的标准偏差为5.273×10-3,其变异系数为1.25%。
The optimum extraction conditions of rutin from asparagus were investigated by using orthogonal design. Quantitative analysis of rutin content was achieved by spectrophotometry and qualitative analysis of rutin was achieved by ultra violet spectro photometry. The result shows that 75%, 25:1 of 75% ethanol to asparagus extraction for 5h was the optimal condition. The extraction rate was 78.7%. The average recovery rate was 97.83%. The standard deviation was 5.273×10^-3and the variation coefficient was 1.25%.
