摘要 Caspase是一组具有相似氨基酸序列、空间结构和底物特异性的半胱氨酸蛋白酶,由其引发的级联反应是细胞凋亡过程中的中心环节。1型、2型糖尿病均以胰岛β细胞凋亡为特征。T淋巴细胞,尤其是CD4+辅助性T细胞通过分泌干扰素-γ、肿瘤坏死因子-α、白细胞介素-1β等细胞因子激活cas-pase,介导了胰岛β细胞凋亡和1型糖尿病的发生。在2型糖尿病中,胰岛淀粉样变、高血糖、高游离脂肪酸等也可通过激活caspase导致β细胞凋亡。Caspase参与了1型、2型糖尿病的发生、发展。 Caspases are a family of eysteine proteases that share the homology of amino acids sequence, spacial structure and substrates specificity. The cascade triggered by easpase family members play a key role in cell apoptosis. It has been suggested that type 1 and type 2 diabetes are characterized by pancreatic islet β cell apoptosis. In type 1 diabetes, T lymphocyte, especially CD4^+ Th produce eytokines, such as interferon-γ, tumor necrosis factor-α, and interleukin-1β. Interferon-γ and (or) tumor necrosis faetor-α plus interleukin-1β induce β cell apoptosis via the activation of easpase. In type 2 diabetes, the elevated levels of islet amyloid, glucose and free fatty acids cause β cell apoptosis through the same pathway. The members of easpase family take part in the onset and development of diabetes.
