MALDI imaging mass spectrometry on biological tissue sections:the new progress

摘要 基质辅助激光解吸电离质谱成像技术分析组织切片已成为临床质谱学的一个新领域。该技术通过研究生物组织产生的“二维离子密度图像”,可以快速评估完整组织中蛋白质、药物及其代谢产物的空间分布。利用质谱成像技术研究阿尔茨海默病、神经胶质瘤、肺癌和前列腺癌等组织取得一定进展,本文对质谱成像原理、方法学及相关应用进行了综述。 MALDI MS imaging for tissue section analysis has opened a new area in clinical mass spectrometry. It allows ion mass spectra to be obtained from intact tissues, forming "two-dimensional ion density maps". It enables the investigator to detect the spatial distribution of proteins, drugs and their metabolites in intact tissues. Certain progress has been made in studying the biopsies of Alzheimer disease, glioma, lung cancer and prostate tumor by imaging mass spectrometry. Principle of imaging mass spectrometry, methodology and its applications to biological tissues are reviewed.
