福辛普利对大鼠肾小球系膜细胞转化生长因子-β_1蛋白及其mRNA表达的影响 被引量:7

摘要 目的观察血管紧张素酶抑制剂(ACEI)—福辛普利(FOS)对脂多糖(LPS)诱导大鼠肾小球系膜细胞(GMC)转化生长因子-β(TGF-β1)蛋白及mRNA表达的影响,探讨FOS延缓肾小球硬化机制。方法按经典方法体外培养大鼠GMC,转种培养后分为对照组、LPS组、LPS+FOS组。采用ELISA法测定GMC培养上清6、12、24 h TGF-β1蛋白水平,荧光半定量RT-PCR法检测TGF-β1mRNA表达。结果LPS诱导组GMC分泌TGF-β1蛋白及mRNA表达均高于对照组,LPS+FOS组GMC分泌TGF-β1及mR-NA表达较LPS组明显下降。结论FOS从蛋白和mRNA两个水平均对体外培养大鼠GMC产生TGF-β1有明显抑制作用,提示FOS抑制GMC产生TGF-β1可能是延缓肾小球硬化的重要作用机制之一。 Objective To observe the effects of fosinopril (FOS), a new generation angiotensin- converting enzyme inhibitor (ACEI) ,on protein and mRNA expression of transforming growth factor-β1 (TGF-β1) of rat glomerular mesangial cell(GMC) induced by lipopolysaccharide(LPS) ; to demonstrate the preventive mechanism against glomerular sclerosis by applying FOS. Methods The cultured GMC in classic way were divided into 3 groups: control group; LPS group; LPS + FOS group. TGF-β1 concentration in GMC superrlatant fluid was detected by ELISA; TGF-β1 mRNA expression was determined by semiquantitative real- time RT- PCR. Results LPS group was obviously higher than control groups in TGF-β1 secretion and mRNA expression, while LPS + FOS group decreased distinctively in TGF-β1 secretion and mRNA expression compared with LPS group. Conclusions FOS has obviously inhibited on TGF-β1 expression of rat GMC both at protein level and mRNA level, which reveals that it may be an important mechanism by FOS on restraining the development of glomerulosclerosis.
