摘要
目的用高效液相色谱法测定桃核承气汤中游离型及结合型芦荟大黄素、大黄酸、大黄素、大黄酚及苦杏仁苷的含量。方法采用C18柱,蒽醌类成分色谱条件为:流动相:甲醇-0.1%磷酸(82∶18,v:v);流速:1.0ml·min-1;柱温:25℃;紫外检测波长:230nm。苦杏仁苷色谱条件为:流动相:乙腈-水(15:85,v∶v);流速:1.0ml·min-1;柱温:25℃;紫外检测波长:191nm。结果桃核承气汤中芦荟大黄素、大黄酸、大黄素、大黄酚、苦杏仁苷的平均加样回收率分别为:99.0%、99.5%、102.4%、103.4%、101.6%;线性范围分别为:0.0104~0.2600μg、0.0576~1.4400μg、0.0096~0.2400μg、0.0117~0.2925μg、0.112~2.800μg。结论本法准确可靠,重现性好,结果稳定,适合于桃核承气汤中蒽醌类成分及苦杏仁苷含量的分析。
Objective To determine the free anthraquinones such as aloe-emodin,rhein,emodin,chrysophanol and their glycosides and amygdalin in "Taohe Chengqi Decoction" by high performance liquid chromatography (HPLC), Methods The analytical column was Eclipse XDB-C18 column (5 μm,4.6×150mm).The mobile phase of anthraquinones was composed of methanol -0.1% phosphoric acid (82:18,v:v) with flow rate of 1.0 ml·min^-1 and column temperature of 25℃ and UV detection wave-length of 230 nm. The mobile phase of amygdalin was composed of acetonitrile-water (15:85, v:v) with flow rate of 1.0ml·min^-1 and column temperature of 250C and UV detection wavelength of 191nm. Results The average recoveries of aloe-emodin,rhein, emodin, chrysophanol and amygdalin were 99.0%,99.5%, 102.4%, 103.4% and 101.6% respectively.Their linear ranges were 0.0104~0.2600 μg for aloe-emodin, 0.0576~1.4400 μg for rhein, 0.0096~0.2400 μg for emodin, 0.0117~0.2925 μg for chrysophanol,0.112~2.800 μg for amygdalin,respectively.Conclusions The methods were accurate,repeatable and suitable to determine the contents of anthraquinones and amygdalin in Taohechengqi Decoction.
