应激性肾上腺素对THP-1源性巨噬细胞TGF-β_1与SR-AImRNA表达的影响

摘要 目的探讨心理应激时肾上腺素对THP-1源性巨噬细胞TGF-β1与SR-AImRNA表达的影响。方法不同浓度肾上腺素(1pmol/L^1μmol/L)处理THP-1源性巨噬细胞24h,运用逆转录多聚酶链反应检测其TGF-β1与SR-AImRNA表达。结果1~10nmol/L的肾上腺素对受试基因mRNA的表达与相应对照组比较差异无显著性意义(P>0·05)。而100nmol/L及1μmol/L的肾上腺素下调TGF-β1mRNA水平和上调SR-AImRNA水平,与各自对照组比较差异均有显著性意义(P<0·05)。结论一定浓度的肾上腺素可下调THP-1源性巨噬细胞TGF-β1mRNA水平,上调其SR-AImRNA水平。 Objective To investigate the effect of adrenaline on the expression of SR - AI mRNA and TGF - β1 mRNA in THP - 1 - derived macrophages at the time of psychological stress. Methods THP - 1 - derived macrophages were incubated with various concentrations (1 pM to 1 μM) of adrenaline for 24 hours. Then SR- AI and TGF- β1 mRNA expression were determined by RT - PCR. Results RT - PCR analysis showed that different concentrations ( 1 pM to 10 nM) of adrenaline had no significant effect on these genes expression ( P 〉 0, 05 ). Meanwhile, compared with control group ( without adrenaline ), adrenaline of higher concentrations (100 nM, 1 μM ) significantly decreased TGF - β1 mRNA expression and significantly increased SR - AI mRNA expression ( P 〈 0. 05 ). Conclusion A certain concentration of adrenaline can down - regulate TGF - β1 mRNA expression and up - regulate SR - AI mRNA expression in THP - 1 macrophages.
